1 · Indicator organisms: coliforms, Enterobacteriaceae, E. coli · easy

Coliforms are defined as Gram-negative non-spore-forming rods that:

  1. Ferment lactose with acid and gas within 48 h at 30–37 °C
  2. Ferment glucose but never lactose
  3. Form heat-resistant spores that survive pasteurisation at 72 °C
  4. Grow only above 55 °C
Answer and working

A. Ferment lactose with acid and gas within 48 h at 30–37 °C

Lactose fermentation with acid and gas is the defining trait; Enterobacteriaceae are defined by glucose fermentation.

2 · Indicator organisms: coliforms, Enterobacteriaceae, E. coli · medium

Coliform counts in raw milk mainly reflect:

  1. Hygiene of milking and equipment
  2. Antibiotic residues
  3. Efficiency of pasteurisation
  4. Shelf life of the packaged product
Answer and working

A. Hygiene of milking and equipment

In raw milk they are expected and indicate milking and equipment hygiene.

3 · Standard plate count and other enumeration methods · medium

The most probable number method is especially useful for:

  1. Very high counts in cheese
  2. Low numbers in liquids
  3. Identifying species
  4. Counting dead cells
Answer and working

B. Low numbers in liquids

MPN estimates concentration statistically from positive tubes at several dilutions; it suits low counts.

4 · Biosensors in dairy safety · medium

Surface plasmon resonance detects binding by a change in:

  1. Colony colour
  2. Refractive index at a metal surface
  3. Electric current at an enzyme electrode
  4. Mass on a quartz crystal
Answer and working

B. Refractive index at a metal surface

SPR is an optical label-free method used, for example, for antibiotic residues in milk.

5 · Biosensors in dairy safety · medium

An amperometric biosensor measures:

  1. Potential at zero current
  2. Resonant frequency
  3. Refractive index
  4. Current
Answer and working

D. Current

Amperometric sensors measure current; potentiometric ones measure potential; impedimetric ones measure impedance.

6 · Biosafety levels and good microbiological practice · easy

Salmonella, Listeria monocytogenes and Staphylococcus aureus are handled at:

  1. BSL-4
  2. BSL-2
  3. BSL-3
  4. BSL-1
Answer and working

B. BSL-2

They are risk group 2 agents: disease is possible but treatable and spread is limited.

7 · Biosafety levels and good microbiological practice · medium

A Class II biological safety cabinet protects:

  1. The worker only
  2. The sample only
  3. Neither the worker nor the sample; it only filters exhaust
  4. The worker, the sample and the environment
Answer and working

D. The worker, the sample and the environment

Inward airflow and HEPA-filtered downflow protect all three; Class I does not protect the sample.

Further reading: Wehr and Frank (eds), Standard Methods for the Examination of Dairy Products, 17th ed.

8 · Biosafety levels and good microbiological practice · medium

A Class III biological safety cabinet is:

  1. A sealed, gas-tight glove box
  2. A laminar flow hood that blows air at the worker
  3. A refrigerator
  4. An open bench with a fan
Answer and working

A. A sealed, gas-tight glove box

Class III cabinets provide maximum containment for the most dangerous agents.
