Aspartic acid has pKa values of 2.09 (alpha-carboxyl), 3.86 (side-chain carboxyl) and 9.82 (amino). What is its isoelectric point?

  1. 5.955
  2. 6.840
  3. 2.975
  4. 5.257
Answer and working

C. 2.975

For an acidic amino acid the neutral (zwitterionic) form lies between the two carboxyl dissociations, so pI = (2.09 + 3.86)/2 = 2.975. Averaging the alpha-carboxyl and amino pKa gives 5.955; averaging side chain and amino gives 6.840; averaging all three gives 5.257.

Complete aerobic oxidation of one glucose yields about 30 ATP rather than 32 when cytosolic NADH enters mitochondria by which route?

  1. The glycerol-3-phosphate shuttle
  2. The malate-aspartate shuttle
  3. The carnitine shuttle
  4. The citrate-pyruvate shuttle
Answer and working

A. The glycerol-3-phosphate shuttle

The glycerol-3-phosphate shuttle passes electrons to FAD, giving 1.5 ATP per cytosolic NADH instead of 2.5, so the two glycolytic NADH give 2 fewer ATP. The malate-aspartate shuttle preserves NADH, giving 32. The carnitine shuttle moves acyl groups.

Cyanide from wilted or regrowing sorghum kills ruminants mainly by inhibiting which enzyme?

  1. Aconitase
  2. Cytochrome c oxidase
  3. ATP synthase
  4. Succinate dehydrogenase
Answer and working

B. Cytochrome c oxidase

Cyanide binds ferric iron in cytochrome a3 of complex IV, halting electron transport; venous blood stays bright red because tissues cannot use oxygen. Aconitase is blocked by fluorocitrate from fluoroacetate; ATP synthase by oligomycin.

4 · Vitamins: biochemical roles · easy

Which enzyme requires biotin?

  1. Pyruvate dehydrogenase
  2. Propionyl-CoA carboxylase
  3. Transketolase
  4. Glutamate dehydrogenase
Answer and working

B. Propionyl-CoA carboxylase

Biotin is the carboxyl carrier for carboxylases: pyruvate, acetyl-CoA and propionyl-CoA carboxylase. Pyruvate dehydrogenase and transketolase use TPP; glutamate dehydrogenase uses NAD(P).

Why is niacin a dietary requirement for cats, even when tryptophan intake is high?

  1. Cats lack the NAD+ binding site on their dehydrogenases and so need niacin as a cofactor
  2. Cats cannot absorb tryptophan
  3. Cats excrete all dietary tryptophan unchanged in the urine before it can be metabolised
  4. High picolinic carboxylase activity diverts tryptophan metabolites away from NAD
Answer and working

D. High picolinic carboxylase activity diverts tryptophan metabolites away from NAD

In most mammals part of tryptophan is converted to niacin, but in cats picolinic carboxylase is so active that almost none is. They absorb tryptophan normally and use NAD+ like other species.

Total serum calcium in a cow is reported as 10 mg/dL. What is this in mmol/L (atomic mass of calcium 40.08)?

  1. 1.25 mmol/L
  2. 0.25 mmol/L
  3. 5.0 mmol/L
  4. 2.50 mmol/L
Answer and working

D. 2.50 mmol/L

10 mg/dL = 100 mg/L; 100 / 40.08 = 2.50 mmol/L. 0.25 forgets the dL-to-L factor of 10; 5.0 is the value in mEq/L (calcium is divalent, so mEq/L = 2 x mmol/L); 1.25 halves the correct value instead of doubling it.

7 · Hormones · hard

In high-yielding dairy cows in early lactation, the somatotropic axis is described as 'uncoupled'. What does this mean?

  1. Growth hormone is high but hepatic growth hormone receptors and IGF-1 are low
  2. Growth hormone and IGF-1 are both high
  3. Insulin is high and growth hormone is low
  4. IGF-1 is high while growth hormone is undetectable
Answer and working

A. Growth hormone is high but hepatic growth hormone receptors and IGF-1 are low

Reduced hepatic GH receptor expression means high GH fails to stimulate IGF-1, so IGF-1 does not feed back to lower GH. High GH and low insulin favour lipolysis and direct glucose to the udder (homeorhesis).

When cellular energy falls, AMP-activated protein kinase (AMPK) phosphorylates and inhibits which enzyme, reducing fatty acid synthesis?

  1. Carnitine palmitoyltransferase I
  2. Hormone-sensitive lipase
  3. Acetyl-CoA carboxylase
  4. Pyruvate kinase
Answer and working

C. Acetyl-CoA carboxylase

AMPK inactivates acetyl-CoA carboxylase; malonyl-CoA falls, releasing inhibition of CPT-I so fatty acid oxidation rises. It does not inhibit CPT-I itself.

9 · Ruminant metabolism: volatile fatty acids, gluconeogenesis and ketogenesis · medium

Which pair of vitamins is required to convert propionate to succinyl-CoA?

  1. Biotin and vitamin B12
  2. Thiamine and riboflavin
  3. Folate and vitamin B6
  4. Niacin and pantothenate only
Answer and working

A. Biotin and vitamin B12

Biotin serves propionyl-CoA carboxylase and adenosylcobalamin (B12) serves methylmalonyl-CoA mutase. Coenzyme A (from pantothenate) is also involved, but the pathway-specific vitamins are biotin and B12.

10 · Ruminant metabolism: volatile fatty acids, gluconeogenesis and ketogenesis · hard

If a cow absorbed 30 mol of propionate in a day and all of it were converted to glucose, what is the theoretical maximum glucose produced (molar mass 180.16 g/mol)?

  1. About 5.4 kg (30 mol)
  2. About 2.7 kg (15 mol)
  3. About 1.8 kg (10 mol)
  4. About 1.35 kg (7.5 mol)
Answer and working

B. About 2.7 kg (15 mol)

Two three-carbon propionate molecules make one six-carbon glucose: 30/2 = 15 mol x 180.16 g/mol = 2702 g, about 2.7 kg. 30 mol assumes one glucose per propionate; 10 mol divides by 3; 7.5 mol divides by 4.
